Overview
Fungal culture inoculates specimens to media such as Sabouraud dextrose agar (± selective agents) and incubates for extended periods to recover fungi.
Clinical Importance
Diagnose superficial and invasive fungal infections

Specimen Requirements
- Specimen: site-appropriate clinical material collected aseptically per institutional protocol
- Container: sterile container or transport medium validated for the ordered test
- Handling: transport promptly at the temperature specified for the method; avoid refrigeration when anaerobes or certain pathogens are suspected unless SOP directs otherwise
- Labeling: two patient identifiers, collection time/date, and anatomic site
- Stability: organism viability and antigen/nucleic acid stability are method-specific; do not exceed local time limits
Analytical Method
Specimens are plated to fungal media and incubated (often at 25–30 °C and/or 35–37 °C per protocol) for days to weeks. Yeasts may be identified rapidly by MALDI-TOF; molds require macroscopic/microscopic morphology and, increasingly, molecular methods.

- Blood culture systems with fungal bottles when fungemia is sought

- Manual mycology culture, tease mounts, and lactophenol cotton blue microscopy
Reference Values
Microbiology results are interpreted against organism identification, specimen quality, clinical context, and current CLSI and/or EUCAST interpretive criteria (including CLSI M100 for susceptibility where adopted). Reporting conventions and critical-notification lists are institution-defined; verify against the performing laboratory SOP and current guideline editions.
Expected: no fungi recovered from sterile sites. Recovery from nonsterile sites requires clinical correlation (colonization vs infection).

- No growth; does not exclude infection (prior therapy, sampling error, dimorphic fungi needing extended incubation)
